This antibody recognizes a protein of 83 kDa, which is identified as Factor XIIIa. Anti-factor XIIIa has been found to be useful in differentiating between dermatofibroma (almost all cases are positive), dermatofibrosarcoma protuberans (-/ ) and desmoplastic malignant melanoma (-). Frataxin is a nuclear-encoded mitochondrial protein implicated in Friedreich's ataxia (FRDA), a human autosomal recessive neurodegenerative disease. Lack of Frataxin causes iron to accumulate in the mitochondrial matrix suggesting that Frataxin is involved in mitochondrial iron homeostasis and possibly in iron transport. This MAb recognizes a protein of 67 kDa, which is identified as glutamic acid decarboxylase 1 (GDA1). There are two forms of glutamic acid decarboxylases (GADs) that are found in the brain: GAD65 (also known as GAD2) and GAD67 (also known as GAD1. GAD65 and GAD67 are members of the group II decarboxylase family of proteins and are responsible for catalyzing the rate-limiting step in the production of GABA (ÃŽ³-aminobutyric acid) from L-glutamic acid. Although both GAD s are found in the brain, GAD65 localizes to synaptic vesicle membranes in nerve terminals, while GAD67 is distributed throughout the cell. GAD67 is responsible for the basal levels of GABA synthesis. In the case of a heightened demand for GABA in neurotransmission, GAD65 will transiently activate to assist in GABA production. This antibody recognizes a protein of 39 kDa, which is identified as Aurora kinase B (or Aurora B). Aurora B is a serine/threonine protein kinase and component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. It is important for accurate chromosome segregation, cytokinesis, protein localization to the centromere and kinetochore, correct microtubule-kinetochore attachment, and regulation of the mitotic checkpoint. Aurora B forms a tight complex with inner centrosome protein and survivin. Inactivation of any of these proteins causes similar defects in chromosome segregation. Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained. Myoglobin is a cytosolic oxygen-binding protein responsible for the storage and diffusion of oxygen within myocytes. Expression of myoglobin is highest in skeletal and cardiac muscle. Recognizes a protein of 40 kDa, identified as CD7 (also known as gp40, Leu9). CD7 is a member of the immunoglobulin gene superfamily. This antibody recognizes CK1, CK5, CK10 and CK14. In normal epithelia, it stains stratified epithelia, myoepithelial cells and basal cells in the prostate gland and bronchi. This MAb shows no reactivity with hepatocytes, pancreatic acinar cells, proximal renal tubules, or endometrial glands; there is no reactivity with cells derived from simple epithelia. Mesenchymal tumors, lymphomas, melanomas, neural tumors, and neuroendocrine tumors are negative with this antibody.
